The formulation of tablets of acetylsalicylic acid for aqueous enteric coating
Monday, September 14th, 2009The objective of this study was to determine which combination of excipients would result in a tablet core which is suitable for use in the process of aqueous enteric coating layer. A relatively simple formulation of microcrystalline cellulose (MCC) and partly pregelatinized starch (PGS-P) were found to provide the required properties. MCC in the formulation provided the compaction necessary to produce a pill that resist mechanical stress of the film coating process. P-PGS provides dissolution characteristics and is responsible for the characteristics of stability in this sensitive to moisture, the application gastro-resistant. It was also found that the P-PGS could be used to reduce the harmful effects of superdisintegrants in formulations.
In recent years, aspirin (ASA, also known as aspirin) was prescribed a number of indications. Besides its use as an analgesic, anti-inflammatory and antipyretic agent, ASA is not indicated for use in the prevention and treatment of heart disease and stroke. Other studies are currently investigating the potential of SAA to stimulate the immune system, cognitive disorders and a reduced risk of colon cancer and ovarian cancer. A low daily dose, 75-81 mg, is commonly used in the preventive treatment of ASA. Historically, the SAA has been considered a potential gastric irritation (1) and studies have shown that the incidence of gastric intestinal side effects may increase with regular use of gastro-resistant (2) of the tablet is highly desirable to avoid stomach discomfort or irritation in patients receiving ASA treatment daily.
Aspirin is a medicine sensitive to moisture and can be hydrolyzed to acetic acid and salicylic acid when exposed to high humidity and high temperatures (3). As the coating process is subject to two tablets of ASA high temperatures and humidity, it is important that the formulation is resistant to the interaction of moisture. Mitrevej Hollenbeck and found that a field is generated around the hydrophilic ASA crystals under conditions of high humidity and that, in combination with certain hydrophilic ASA disintegration, condensation in the vicinity of the crystal can occur ASA (4). The disintegrants were studied starch glycolate (SSG), croscarmellose sodium (CCS), crospovidone and colloidal silica. During the process of aqueous film coating, Faroongsarng and Peck found that the depth of water penetration into the tablet core may be directly related to the concentration and type of disintegrant used in the formula (5). Further work studying Taani Bashar Al-aqueous coating for tablets ASA confirmed that the penetration of moisture during the coating process is not only the drafting process, but could be directly linked to the stability of tablets coated with SAA final (6).
A review of the ingredients in five products purchased commercially ASA considered that, in most cases, the main listed excipients are microcrystalline cellulose (MCC) and some form of starch. The use of additional excipients, including disintegrants (such as CCS and SSG), lubricants and Glide varied. The five products are packaged in paper, sealed high density polyethylene (HDPE), three of which contained carbon / silica desiccant packets. The objective of this study was to determine which combination of excipients, commercial products ASA, lead to a tablet core which is suitable for use in a process aqueous enteric film coating. Ideally, enteric coated tablets which exhibit excellent storage stability under accelerated without the fuse more (and more expensive) precautions in packaging as desiccant packets for other specialist packaging materials.
Machinery and equipment
Aspirin 1040 (Aspirin USP mesh of 40 crystals, Rhodia, Cranbury, New Jersey, USA) was used as an active ingredient. The excipients used in the study were partially pregelatinized starch (PGS-P) (Starch 1500, Colorcon, West Point, Pennsylvania, USA), MCC (Emcocel 50M, Penwest, Patterson, New York, USA), SSG (Explotab, Penwest), CCS (Ac-Di-Sol, FMC, Princeton, New Jersey, USA) and stearic acid NF (purified grade powder plant Oleotec Ltd., London, UK). The packaging material used was 85 mL aluminum sealed bottles of high density polyethylene (Drug Plastics & Glass Co., Boyertown, Pennsylvania, USA) and desiccant packets (3964, Süd-Chemie Performance Packaging, Belen, New Mexico, USA). The materials used were an aqueous layer enteric coating system (Opadry II), both manufactured by Colorcon.
Dry ingredients mixed in a double 16-shell blender (Patterson-Kelley Co., East Stroudsberg, Pennsylvania, USA). The tablets were compressed to 10 station rotary press instrumented Piccola (Riva, Buenos Aires, Argentina). Tablet strength was measured using a tester Multicheck (ERWEKA, Milford, Connecticut, USA). A team of 15-inch vented coating pan (Labcoat II, O'Hara Technologies, Toronto, Canada) was used to apply coatings. A dissolution test station (VK 7010, a device that Vankel, Cary, North Carolina, USA) was used for the release of drug testing. An HPLC (high performance liquid chromatography system (Alliance 2690, Waters Corp., Milford, Mass.) was used to determine the concentration of salicylic acid free.
Methods
Preparation of the mixture and Tablet
Six formulations (see Table I), consisting of constant levels of AAS and lubricants and different levels of CMC, PGS-P, CCS and SSG each mixture is dried for 15 minutes in the mixer jacket. The lot size of each mixture is 5kg. Each of the six mixture was compressed in the 10-station rotary tablet press at 7.0 mm standard concave tool. The target tablet weight was 162.0 mg and the compaction force was adjusted to produce tablets with a breaking strength of 6.0-7.0 kp. The coating of tablets was performed in 15-inch side-vented pan fitted with a gas pistol. The bread was 3 kg load. A subcoat of Opadry II dispersed in water (15% w / w) was applied to obtain a theoretical gain of 2% weight Tablet tablets of six lots. Applications subcoat was immediately followed by a layer consisting of enteric Sureteric dispersed in water (15% w / w) and applied to obtain a theoretical weight gain of 10%. A coat of Opadry II dispersed in water (15% w / w) was then applied to the tables to weight gain of 2%. The six tests of coating is achieved using the same process temperature recommended spray rates, and operating conditions. In general, using a subcoat under the enteric coating is optional and depends largely on the quality of the tablet core. About six batches contained different ingredients, a subcoat was applied to the six batches so that the enteric coating would be affected by minor changes in the surface of the tablet. Wear a topcoat which is optional, but many commercial products that have a top coat of color applied to the base.
Dissolution of the analysis of salicylic acid free
The dissolution tests and without salicylic acid were performed according to the USP 23 monograph for tablets ASA. Film-coated tablets were analyzed by the USP 23 monograph for delayed-release tablets ASA.
Tablet Hardness Testing
The uncoated tablets were tested for resistance to diametrical fracture before and after storage in accelerated conditions. The average result was reported for 20 tablets tested.
The packaging and stability
Samples of the uncoated tablets of each formulation were packaged in bottles of high density polyethylene (120 tablets per bottle). Film-coated tablets of each formulation were packaged the same way: a set of samples has been packaged without desiccant, a second set of samples was packaged with a packet of desiccant in each bottle. All bottles are induction (aluminum) sealed and stored under accelerated conditions, 40 degrees Celcius/75% relative humidity (RH) for 3 months.
Results and discussion
ASA Uncoated Tablets
Tests conducted in an acetate buffer solution (pH = 4.5) revealed that only one lot containing only MCC as the vehicle could not achieve 80% drug release within 20 min. The results after the dissolution of storage in accelerated conditions showed little change in initial tests.
More significant are the results of strength tablet after exposure to accelerated temperature and humidity conditions. The tablets containing ASA and MCC has lost only 8.57% of the hardness of the tablet while tablets containing CM-P PGS combination showed the lowest decrease in hardness of the tablet, with a loss 3.0%. The use of these technologies or any other SGS in association with MCC caused a loss of more than 36.3% in the mechanical strength of tablets. Interestingly, when the same levels of CCS and SSG were used in the tables combined PGS P, and CMC, the loss of hardness of the tablet is less deep. By comparing the levels of salicylic acid, not lying in the initial time point and after 3 months to 40 degrees Celcius/75% relative humidity, the results show a trend similar to the results of the hardness of tablets.
The USP limit for free salicylic acid in film-coated tablets ASA is only 0.3%. After 3 months in accelerated conditions, the tablets containing only CMC or MCC as an excipient, or CCS or ECG showed significantly higher levels of salicylic acid free and does not meet the requirements of USP. The CM-P-PGS combination showed almost no degradation of the ASA, with a time of storage under adverse conditions, and increased free salicylic acid is negligible.
It has been demonstrated that the P-GSP used in this study has a lower propensity to absorb moisture than any of SSG and CCS or less moisture will be based on a shelf under conditions of high humidity ( 7). This may partly explain the positive impact they have had with their use in this formulation. The data also suggest that the P-PGS may be able to trap and retain moisture in the formulation, thus delaying moisture interaction with the SAA.
The first results of ASA film-coated tablets
After coating, the tablets of all formulations have a good appearance. None of these tablets showed signs of defects, either during or after the test coating. Tablets of all batches passed the test phase of the acid solution without possibility of release of the ASA, after 2 hours in 0.1N HCl During the test phase buffer (pH = 6.80, the results of the dissolution of uncoated tablets, the tablets containing only MCC and ASA not only meet USP specifications for no less than 80% ASA Released in 90 min. In fact, five other formulations ASA output reaches 80% within 20 min.
Results of stability-coated tablets
After 3 months of storage at 40 degrees Celcius/75% humidity, some of the tablets containing SSG SAC or softening of the exposed layer of the film and the adhesion of the tablets with each other inside the cylinder High density polyethylene. This occurred in samples which were packed with and without desiccant packets. The tablets show signs of defects at this point were considered as failures of stability.
The results without salicylic acid-coated tablets are very similar to results obtained for the uncoated. The USP limit for free salicylic acid tablets coated ASA, 3.0%, exceeding the specification coated tablet. After 3 months in accelerated conditions, the tablets containing only MCC as the vehicle was higher, but acceptable, free salicylic acid levels. The combination of CCS and SSG MCC resulted in substantial increases to more than 5.0% of salicylic acid free in general, so it does not meet the requirements of USP. Again, the most acceptable results were observed for tablets containing MCC and PGS-P and excipients, which show no increase of free salicylic acid was used as desiccant and only an increase of 0, 91% when packaged without desiccant. The addition of P-PGS substantially reduces the amount of degradation AAA tablets containing CMC combined with SSG or SCC, which have unacceptable levels of salicylic acid free.
Interestingly, the addition of desiccant packs of bottles is not sufficient to eliminate or even substantially reduce the adverse effects of superdisintegrants. Of the six formulations, tablets containing MCC MCC alone or combined GSP-P-excipient meet the performance requirements desired stability of the beauty, resistance to acids and acceptable levels of salicylic acid free. The formulation with only MCC does not meet the requirements of the dissolution of delaying the release of AAA in the buffer, either initially or after storage for 3 months under accelerated conditions. The tablets containing CMC-P-PGS This combination of excellent results in the dissolution of delayed release initially and after 3 months to 40 degrees Celcius/75% relative humidity.
Conclusions
The results obtained in this study gave a relatively simple formulation ASA using a combination of MCC and PGS-P primary and excipients. MCC in the formulation requires the compaction necessary to produce a tablet to withstand the mechanical stress of the film coating process. The starch solution provides the functionality needed in this moisture-sensitive, application of the enteric film-coated tablets. The formula without the use of additional superdisintegrants well suited to aqueous film coating process and the final film-coated tablets should not be necessary to use any special packing materials. It was also found that the P-PGS could be used to reduce the harmful effects of superdisintegrants in formulations. It would also reduce the costs of raw materials. The next phase of this study will focus on optimizing the necessary levels of enteric coating and the extension of the enteric coating process.

A softer coat Goldendoodle is, of course, …… But Goldendoodle, Goldendoodle with an easy to maintain and Goldendoodle much like the Golden Retriever breed. There are, however, smooth coat Goldendoodle differences.The may seem to have a similar appearance to that of a purebred dog for gold, but you will notice that the legs may be long, as the poodle ….. your mouth may be as thin as the poodle …….. May which have fluff on the ears and neck and, of course, their bone structure can be smaller, like Poodle.Since Goldendoodles inception in 1999, we now know that there are three layers for the types of dog Goldendoodle: A. ShaggyB. CurlyC. SmoothWe soft layer obtained our first time in 2007 when we decided to set up one of our blue eyes, curly hair Goldendoodles one of our breed Golden Retrievers. "Meeryn" (as shown in photo above) was our first smooth coat Goldendoodle. We intend to bind their parents has been to try to have at least one blue-eyed puppy or a design with multi-colored eyes. This does not occur with this route. It seemed interesting, so it was only 1 smooth coat doodle is born in a litter of six. The rest were men who had their fur. We are pleased that the Golden Retriever temperament is established in the offspring. Some farmers have asked how can you tell if your Goldendoodle puppy will be smooth, curly hair or fur and our response has always been to wait until the pup is closer to six weeks. There are specific signs to look to go with three types of coat. An experienced breeder will know with time if they have managed to combine a particular way Goldendoodles will have one or more of each type of housing. The inexperienced breeder must wait until the puppy to come to six or eight weeks age.With soft layer, their body hair seems to be taken lightly but there will be no test on the face of the usual signs of a bearded face is formed. All puppies Goldendoodle except for the soft layer, will have what we call upside "down V-shaped" with a layer of facial hair between the eyes nose.
